Wnt信号传导路径,从Wnt蛋白配体与Frizzled家族的受体结合开始,通过细胞表面受体,将生物信号经由细胞内的Dishevelled,因而将信号传递到细胞中。
Wnt蛋白是分泌的型态发生因子,是各种发育过程(包括细胞命运决定、细胞增殖和细胞死亡)所必须的,与Frizzled受体结合后,触发了三个途径: 经典途径、平面细胞极性途径和Wnt/Ca2+途径,在经典途径中,主要效应物β-catenin进入细胞核并促进Wnt调控的基因表达,平面细胞极性途径通过启动RhoA和Rac促进细胞骨架重塑和细胞运动,Wnt/Ca2+途径通过增加瞬时Ca2+水平以及随后启动PKC、CAMKII和Calcineurin来介导多种功能,Wnt途径不仅促进包括顶/基极极性和神经管构图等胚胎发生的方面,而且还诱导肿瘤发生,最近还报导了Wnt信号传导调节干细胞分化。
